Semaglutide belongs to a class of medications called GLP-1 receptor agonists. It is prescribed as part of a supervised weight management program alongside changes to nutrition and activity, not as a standalone treatment.

How it works

GLP-1 is a hormone the body releases after eating. It signals fullness to the brain, slows how quickly the stomach empties, and supports the pancreas in releasing insulin when blood sugar rises.

Semaglutide mimics that signal and stays active far longer than the natural hormone, which is why it is dosed weekly rather than daily.

Tolerability

Side effects

The most commonly reported side effects are gastrointestinal: nausea, vomiting, diarrhea, constipation and abdominal pain. These are often most noticeable after a dose increase.

Less common but serious reported effects include pancreatitis, gallbladder disease, kidney injury related to dehydration, and allergic reactions. Contact your provider promptly about severe or persistent abdominal pain.

Boxed warning. In rodent studies, semaglutide caused thyroid C-cell tumors. Whether it causes these tumors in humans, including medullary thyroid carcinoma, has not been determined. It should not be used by anyone with a personal or family history of medullary thyroid carcinoma or Multiple Endocrine Neoplasia syndrome type 2.
